Development study of the inflammatory process during colibacillosis enteritis in broiler chickens: anatomo-pathology and clinical repercussions in poultry farming

| dc.description | La colibacillose est un problème grave qui menace le secteur avicole dans l'est de l'Algérie. Escherichia coli (E. coli) est l'espèce la plus pathogène chez les poulets de chair, entraînant de lourdes pertes économiques dans la filière avicole. L'infection peut entraîner une mortalité élevée, une diminution de la production et de la qualité de la viande et une augmentation des coûts vétérinaires, en plus d'un problème de résistance aux antibiotiques et un risque sur la santé publique. Escherichia coli pathogène aviaire (APEC), appartenant à la famille des E. coli extra-intestinaux, provoque différents types d'infections, notamment : septicémie, aérosacculite et entérite. Dans cette étude, notre objectif était de mettre en évidence les lésions histopathologiques causées par l'infection à Escherichia coli pathogène aviaire (APEC), ainsi que l'évolution du score lésionnel de l'intestin au cours de l'entérite colibacillaire. Pour cela cent cinquante échantillons ont été collectés dans quinze élevages avicoles dans la région de Constantine (Est algérien), où les poulets de chair présentaient de nombreux signes cliniques, tels que diarrhée aqueuse, déshydratation et taux de mortalité élevé ; en outre, l’autopsie a révélé un gonflement intestinal avec la présence des fluides et des taches de mucus. D'autre part, afin d'étudier l'effet immunosuppresseur de l'APEC sur la bourse de Fabricius, cinquante et un échantillons de bourse ont été prélevés sur des poulets présentant une coli septicémie, pour une étude histologique. L'infection par l'APEC a été confirmée par un test au rouge Congo, et le nombre d'Escherichia coli a été estimé en utilisant la méthode de Miles et Misra. Ensuite, les échantillons ont été fixés dans du formol 10⸓ et soumis à un laboratoire de pathologie pour une étude histologique selon le protocole de Luna. Nos résultats ont révélé que sur cent cinquante échantillons, cent trente-sept (91,3⸓) étaient positifs au rouge Congo, avec des concentrations d'E. coli allant de 5*107 à 2*109. Une analyse minutieuse de 236 lames a révélé des lésions dégénératives intestinales induites par une infection à APEC. Ces lésions se caractérisent par une congestion, un œdème, une hypertrophie de la lamina propria avec infiltration de cellules inflammatoires, une atrophie des cryptes, ainsi qu'une desquamation et une nécrose des villosités et de l'épithélium des cryptes. Ces lésions ont été observées principalement au niveau du duodénum. La bourse de Fabricius présentait également des modifications pathologiques importantes, notamment : des structures semblables à des glandes dans les follicules avec un épithélium ondulé, un œdème, une hypertrophie, une hémorragie et une infiltration de cellules inflammatoires dans les régions interfolliculaires, des vacuolisations et une nécrose dans les zones médullaires, un excès de tissu conjonctif fibreux s'était formé, des vacuolisations de tailles variables dans les plis de la lamina propria, des zones contenant des débris nécrotiques accumulés dans la médullaire des follicules, une déplétion lymphocytaire et une atrophie des follicules. En conclusion, l'infection par APEC induit d'importantes modifications histologiques intestinales, principalement observées au niveau du duodénum. La bourse de Fabricius présente des altérations significatives. Ces modifications ressemblent fortement à celles généralement observées lors d'infection par le virus de la bursite infectieuse. Les similitudes morphologiques sont frappantes, suggérant un impact significatif sur l'immunité humorale. | |
| dc.description.abstract | Colibacillosis constitutes a serious danger to poultry production in eastern Algeria. Escherichia coli, particularly the avian pathogenic strain (APEC), is the primary pathogen affecting broiler chickens, causing substantial economic losses through high mortality rates, decreased meat yield and quality, elevated veterinary expenses, antibiotic resistance concerns, and public health risks. APEC, an extraintestinal pathotype, induces various infections including septicemia, airsacculitis, and enteritis. In this study our objective was to highlight histopathological lesions caused by avian pathogenic Escherichia coli (APEC) infection, in addition to the evolution of the lesion score of the intestine during colibacillosis enteritis. This study aimed to characterize the histopathological lesions caused by APEC infection and track the progression of intestinal lesion scores during colibacillosis enteritis. We collected 150 samples from 15 poultry farms in Constantine, eastern Algeria, where broilers exhibited clinical signs such as: dehydration, watery diarrhea, and elevated mortality. Necropsy revealed intestinal distension filled with fluid and mucus flecks. Additionally, to assess APEC's immunosuppressive effects, 51 bursa of Fabricius samples were obtained from chickens with coli septicemia for histological examination. APEC infection was confirmed via a Congo red dye assay, and the number of Escherichia coli was estimated via the Miles and Misra method. APEC infection was confirmed using the Congo red assay, with bacterial quantification performed via the Miles and Misra method. Samples were fixed in 10% formalin and processed for histology following the Luna protocol. Results showed that 137 of 150 samples (91.3%) were Congo red positive, with E. coli counts ranging from 5*10⁷ to 2*10⁹ CFU/ml. Microscopic analysis of 236 slides revealed characteristic intestinal degenerative lesions from APEC infection, including congestion, edema, expanded lamina propria with inflammatory cell infiltration, crypt atrophy, epithelial desquamation and necrosis of villi and crypts, most prominent in the duodenum. The bursa of Fabricius displayed significant pathological changes, such as gland-like follicular structures with corrugated epithelium, interfollicular edema, enlargement, hemorrhage and inflammatory cells infiltration, medullary vacuolization and necrosis, excessive fibrous connective tissue, epithelial plicae vacuolization, necrotic debris accumulation, lymphocytic depletion, and follicular atrophy. To conclude, avian pathogenic Escherichia coli infection triggers substantial histological alterations in the intestine, predominantly in the duodenum. The bursa of Fabricius exhibits marked changes that closely mimic those seen in infectious bursal disease virus infections. These striking morphological similarities indicate a profound effect on humoral immunity. | |
